If I have a web server (runnign IIS 6) that does not have Server Extenions installed, can my users still login to the website using FrontPage (File, Open Site) make changes and then save? Or will they have to do it differently?

Publishing via Frontpage requires Frontpage Server Extensions. If you do not have this, they will need to use FTP. There are also other Frontpage Components that require Frontpage Server Extensions, so you will need to make sure the customers know that these components will not function.
